Motsau covered the 1.609km distance in 4:22, holding off Marco Bucarizza by two seconds in the dash for the line.
Olympic 800m semifinalist Rynardt van Rensburg finished third, a further two seconds behind.
In the women’s race, Diana-Lebo Phalula followed up on her second-place finish at the Spar Women’s Challenge 10km race in Centurion the day before, taking the win in 5:14.
Her twin sister, Lebogang Phalula, was four seconds off the pace in second position.
Meanwhile, former national half-marathon champion Lucky Mohale won the 10km race in 31:33, crossing the line three seconds ahead of Jeffrey Gwebu.
Former steeplechase track specialist Tebogo Masehla won the women’s contest in 40:16, four seconds clear of Mpho Mabuza in another hard-fought battle.
